Molecular identification of scale insect (Eulecanium giganteum) in Hibiscus rosa-sinensis

Hibiscus rosa-sinensis is a widely grown evergreen valuable medicinal, ornamental species planted in India. Scale insects are small herbivorous found on all continents and they serious sap sucking pests of many plants. These scale undetectable due to their tiny size, basic morphology, polyphagous feeding nature. Hence, the management these become concern across globe. To afford prospective solution problem, an accurate, simple, developmental-stage-independent identification method required, hence this study attempted molecular insect using mitochondrial gene Cytochrome Oxidase Subunit I (mtCOI) sequencing. The experiment was carried out by isolating DNA modified CTAB method. Through two or three rounds error-prone PCR followed steady procedure amplify mtCOI region. This region has been used as standard barcode for diverse array taxa. confirmation done sequencing which suggest highest similarities with Eulecanium giganteum. addresses questions biodiversity characterization insects. Further, information obtained provides baseline data future crop improvement programs integrated pest strategies.
